Title: Green Synthesis of Copper Oxide Nanoparticle using Hibiscus rosa-Sinensis leaf extract and its antibiofilm activity against Pseudomonas aeruginosa

Abstract

There is an increasing focus on eco-friendly green routes for the production of metal nanoparticles using biological entities. In this present study the leaf of the plant Hibiscus rosa-Sinensis, was used to investigate their efficiency for synthesizing copper oxide nanoparticles. The synthesized copper oxide nanoparticles (CuO NPs) were confirmed by the change of colour from green to brown due to the mechanism of surface plasmon resonance. The purity of the biologically synthesized copper oxide nanoparticles (NPs) were confirmed by using the characterization techniques such as UV-Vis spectroscopy, Fourier transmission infrared spectroscopy (FT-IR), X-ray diffraction analysis (XRD), Scanninmg electron microscopy (SEM), HR-TEM and Energy dispersive X-ray analysis (EDX). Antibiofilm activity of the CuO NPs was investigated against the strongly biofilm produced microorganism Pseudomonas aeruginosa. The size of the CuO NPs obtained in this study were in the range between 25-40nm. From the study it was predicted that at the highest concentration of CuO NPs treatment (2.2 µg/mL) the biofilm formation of Pseudomonas aeruginosa was completely inhibited showing 97.68%.
